Ancestry.com Officially Retires Y-DNA and mtDNA Testing

Ancestry.com announced today that they are officially retiring five of their sites and/or products as of September 5, 2014. These include Mundia, Genealogy.com, MyCanvas, MyFamily and, of special interest to my readers, the Y-DNA and mtDNA tests (now being called LegacyDNA). The sales of these tests have been halted, effective immediately.
I don't think the news about the retirement of the Y-DNA and mtDNA tests will come as a big surprise to most of us since Ancestry.com has been focusing their resources on the AncestryDNA autosomal DNA test and have been backing away from promoting the direct line tests for some time. As has been noted by many in the genetic genealogy community, it has become increasingly difficult to find the ordering interface for these tests over the last year or more.

I participated in a conference call detailing these changes with bloggers yesterday where we were given an opportunity to ask questions. I asked several about the Y-DNA and mtDNA tests, as follows:

Q: Will the entire Y-DNA and mtDNA site interface be retired? Will you be able to view and contact your matches? 
A: The entire interface will be retired, including the match lists and the ability to contact your matches.

My comments: If you have tested there, I strongly encourage you to contact your matches before September 5th (and so does Ancestry.com) because you will not have another opportunity to do so.  You can download your raw data (CSV file) until September 5th by going to www.DNA.Ancestry.com and upload to other services like Family Tree DNA. FTDNA has a transfer program for Y-DNA tests here. For $19 you will be able to upload your results to the FTDNA site. Here is the description of their transfer product:


This option is available for customers who have 33 or 46-marker Y-DNA results from a third party company that used the Sorenson's laboratory (this includes tests performed by Ancestry, GeneTree, and Sorenson's SMGF). You may use this option to upload your results to the Family Tree DNA database. The $19 fee will provide you with a Family Tree DNA personal page which will allow you to:
  • Join Family Tree DNA projects freely, but you will not receive matches or a haplogroup prediction.
  • Your uploaded results will be available to the administrator and included on the project's public page for comparison with other project members.

For an additional $39 you can get a new kit from FTDNA. If you tested 33 markers at Ancestry.com, then you will have the additional markers filled in to be equivalent to FTDNA's 25 marker test or if you tested at the 46 marker level at Ancestry.com, then you will receive 37 marker results from FTDNA with this additional test. At the very least, I encourage you to add your results to the free Ysearch site (note the conversion needed) and Mitosearch site.

Q: Are there any Y-DNA or mtDNA test still in the queue and, if so, will these be completed or refunded?
A: There are a small number of Y-DNA and mtDNA tests still in the queue. If you are one of these, then you will be receiving an email detailing your options. Ancestry.com encourages you to call their customer service to discuss this. 

Q: What will happen to the stored DNA samples associated with these tests?
A: The stored DNA associated with these tests will be destroyed. 

Q: Can the stored DNA be returned to the testers or their families rather than being destroyed?
A: No.

Q:  Can those stored samples be used to upgrade to an autosomal DNA test before they are destroyed?
A: Ancestry.com does not currently have anything in the works for doing this.

An additional question was posed by another blogger regarding whether the halpogroup designations that are populated on testers' Ancestry.com user trees will remain. They are going to get back to us about this.

Family Tree DNA Announces the March mtDNA Madness Sale - The Benefits of Full Mitochondrial DNA Sequencing

I just received an email from Family Tree DNA announcing a sale on their mtDNA Full Sequence test, starting tomorrow (copied below). This is their lowest price ever and, fortunately, includes upgrades. 

Although mitochondrial DNA is not the first test that I would recommend for a genealogist, in my recent work I have become increasingly interested in full sequence mtDNA testing. The reason for this is that, in some instances, the full sequence allows for geographic specificity, which can potentially open new and valuable avenues of discovery. The lower resolution tests are not often helpful, so if you are going to order a mtDNA test, then the full sequence is the way to go. (With this very reduced price, this is a good time to try it.)

I can't share most of the instances (yet) that it has been useful for my recent research since some were in the course of my work for "Finding Your Roots with Henry Louis Gates, Jr." (scheduled to air Tuesdays starting at the end of September) and others were for private projects, but I can give an example of a situation where mtDNA testing might be helpful for our genealogy research outside of the typically discussed applications. 

I have long been brickwalled on my German Stolebarger line and do not know if Sarah (maiden name unknown), the wife of John Stolebarger, was German like her husband. Since my mother's first cousin is a matrilineal descendant of Sarah's, I asked him to take the mtDNA test for me two years ago (HVR1 + HVR2). The only match was with someone who is brickwalled at his ancestor, also named Sarah from the exact same time and place as my Sarah! So, matching didn't help us. I didn't plan to upgrade for matching purposes since there were no other matches and he only tested at the lower resolution. 

Recently the mtDNA Haplogroup I Project administrator wrote to me and expressed interest in my cousin's rare mtDNA signature, requesting that I consider upgrading to the full sequence. That got me thinking and, with the great timing of this new offer, I now plan to go ahead and upgrade to see if my cousin's unique mutations can  pinpoint a specific geographic region. This may, at least, help to determine if Sarah was German or of completely different ancestral origins. Is it guaranteed to work? No, but I'm ready for the "Hail Mary" play on this one and, anyway, aren't we all accustomed to trying different approaches to our research challenges?

Family Tree DNA Announcements: New Pricing, New Staff and a Discovery


Family Tree DNA made two announcements today.

The first shares the news that the Y-DNA12 test will be permanently offered at $49. The current $39 sale will end on March 31st, so if you want more of these tests order them now.  The basic mtDNA test will soon undergo a similar reduction. Family Tree DNA will also be adding new customer service and IT staff to keep up with the growing demand. This is all great news for the genetic genealogy community.

The second was a press release detailing the exciting scientific discovery that was made through a customer from the FTDNA database thanks to the astute project administrator Bonnie Schrack. I wrote about the genealogical implications of this discovery earlier this month here.

The following email was sent to project administrators (bolding mine):

It is with great satisfaction that we announce a permanent price change and other improvements that will positively impact your projects.

Prices:
Due to the recent upgrade of our state-of-the-art laboratory, coupled with research and development into increasing lab efficiency, we are able to permanently offer the basic Y-DNA12 test for $49 beginning April 1, 2013. The current sale of Y12 at $39 will end at 11:59PM CST, March 31, 2013. We are also working on bringing down the price of the basic mtDNA test as well; we hope to have that accomplished during the first half of this year.


We understand that several projects have a minimum requirement of 37 markers for the Y-DNA test, but we’ve learned through the promotion with the lower price point on the Y-DNA12 last month, that it did not reduce the number of orders at the 37-marker level. Therefore, there was a net gain for the database, not only by increasing the number of members, but also increasing the number of potential upgrades to higher levels.

We hope that you can encourage family and friends who saw the price as a barrier to now come on board. It is our goal to ensure every single person is able to have the "DNA experience," at least at the basic level. Customer Service:

Every month our lab in Houston processes tens of thousands of discrete tests.

  3. Writing the code for new features to be implemented, whether those features are conceived in-house or at the suggestion of our customers.
Obviously, they also have to fix bugs, because as we know - unfortunately - bugs happen. Any of you that have iPhones, iPads or Android apps know that we receive almost daily notifications for updates which most of the time relate to bug fixes. That doesn’t mean we accept all bugs as inevitable—beyond a certain point they’re not acceptable, so we are also taking the necessary steps to improve this situation by hiring additional qualified people. Houston/March 26, 2013 /PRNewswire/ -- Gene By Gene, Ltd., the Houston-based genomics and genetics testing company, announced that a unique DNA sample submitted via National Geographic's Genographic Project to its genetic genealogy subsidiary, Family Tree DNA, led to the discovery that the most recent common ancestor for the Y chromosome lineage tree is potentially as old as 338,000 years.  This new information indicates that the last common ancestor of all modern Y chromosomes is 70 percent older than previously thought.

The surprising findings were published in the report "An African American Paternal Lineage Adds an Extremely Ancient Root to the Human Y Chromosome Phylogenetic Tree" in The American Journal of Human Genetics earlier this month.  The study was conducted by a team of top research scientists, including lead scientist Dr. Michael F. Hammer of the University of Arizona, who currently serves on Gene By Gene's advisory board, and two of the company's staff scientists, Drs.Thomas and Astrid-Maria Krahn.

The DNA sample had originally been submitted to National Geographic's Genographic Project, the world's largest "citizen science" genetic research effort with more than 500,000 public participants to date, and was later transferred to Family Tree DNA's database for genealogical research.  Once in Family Tree DNA's database, long-time project administrator Bonnie Schrack noticed that the sample was very unique and advocated for further testing to be done.

"This whole discovery began, really, with a citizen scientist – someone very similar to our many customers who are interested in learning more about their family roots using one of our genealogy products," said Gene By Gene President Bennett Greenspan.  "While reviewing samples in our database, she recognized that this specific sample was unique and brought it to the attention of our scientists to do further testing.  The results were astounding and show the value of individuals undergoing DNA testing so that we can continue to grow our databases and discover additional critical information about human origins and evolution."

The discovery took place at Family Tree DNA's Genomic Research Center, a CLIA registered lab in Houston which has processed more than 5 million discrete DNA tests from more than 700,000 individuals and organizations, including participants in the Genographic Project.  Drs. Thomas and Astrid-Maria Krahn of Family Tree DNA conducted the company's Walk-Through-Y test on the sample and during the scoring process, quickly realized the unique nature of the sample, given the vast number of mutations.  Following their initial findings, Dr. Hammer and others joined to conduct a formal study, sequencing ~240 kb of the chromosome sample to identify private, derived mutations on this lineage, which has been named A00.

"Our findings indicate that the last common Y chromosome ancestor may have lived long before the first anatomically modern humans appeared in Africa about 195,000 years ago," said Dr. Michael Hammer.  "Furthermore, the sample, which came from an African American man living in South Carolina, matched Y chromosome DNA of males from a very small area in western Cameroon, indicating that the lineage is extremely rare in Africa today, and its presence in the US is likely due to the Atlantic slave trade.  This is a huge discovery for our field and shows the critical role direct-to-consumer DNA testing companies can play in science; this might not have been known otherwise."

Family Tree DNA recently dramatically reduced the price of its basic Y-DNA test by approximately 50%.  By offering the lowest-cost DNA test available on the market today, Gene By Gene and Family Tree DNA are working to eliminate cost as a barrier to individuals introducing themselves to personal genetic and genomic research.  They hope that expanding the pool of DNA samples in their database will lead to future important scientific discoveries.

An Important Update on SMGF from Dr. Tim Janzen


Dr. Tim Janzen generously shared some very important information with me regarding the future of Sorenson Molecular Genealogy Foundation (SMGF). With his kind permission, I have decided to share this timely update with my readers in Tim's own words, as follows:
 

Since early May 2012 it has been officially known that Ancestry.com is purchasing the genetic genealogy company GeneTree. At that same time it was announced that Ancestry.com had also acquired the DNA related assets from the Sorenson Molecular Genealogy Foundation. (See this post for background regarding that announcement.) Exactly how this would impact the SMGF has been quite unclear since the announcement was made. The origin of the SMGF dates back to 1999 when the billionaire James Sorensen and the geneticist Scott Woodward established the foundation. 

The vision of the SMGF and the promises it made to the genetic genealogy community are summarized here. I will quote some of the content from that web page as follows:  

"The Sorenson Molecular Genealogy Foundation is a non-profit organization dedicated to building the world's foremost collection of DNA and corresponding genealogical information. The Foundation is a world leader in DNA research with direct application to genealogy. SMGF was inspired by discussions in 1999 between philanthropist James LeVoy Sorenson and BYU Professor Scott Woodward about using DNA in genealogy. Since that time, SMGF has collected more than 100,000 DNA samples, together with four-generation pedigree charts, from volunteers in more than 100 countries around the world. Y-chromosome DNA results and pedigree charts are available for searching in the Sorenson Database. Y-DNA results and pedigrees help you trace your direct paternal line. Mitochondrial DNA results and pedigree charts are available for searching in the Sorenson Database. Mitochondrial DNA results and pedigrees help you trace your direct maternal line. The Foundation is also conducting research on autosomal DNA, and plans to release the Sorenson Autosomal Database in the near future. Autosomal DNA results and pedigrees can help you trace all family lines. We invite you to participate in the SMGF project and search the Sorenson Database. Help grow the genetic family tree, one branch at a time." 

The SMGF was heavily funded by James Sorenson until his death in 2008. After that time the SMGF appears to have significantly slowed its testing of the samples sent to the foundation and the staff has also been reduced substantially. At its peak the SMGF employed about 40 full-time genealogists working on entering the information contributed on pedigree charts into its integrated genealogy database, which had 2,694,224 unique ancestors in it at the time the database was last updated in March 2011. The SMGF stopped offering free kits from its web site over 2 years ago. Additional information about the SMGF may be found here.

On June 2, 2012, Steve Perkins blogged that he queried the SMGF about the status of their databases. The response he received from the SMFG may be found on his blog here.

In that correspondence, SMGF indicated that they would not be releasing the autosomal DNA data that they have accumulated since the foundation started. As of August 2009 the SMGF had tested 78,568 samples for an average of 68.5 autosomal STRs and had tested 73,394 samples for 13 X chromosome STRs. This information has never been made public. 

I have been heavily involved in collecting samples for the SMGF on a volunteer basis since 2006. I have either directly or indirectly collected about 2000 samples and associated pedigree charts that I have sent to the SMGF. About 85% of those samples were from people of Low German Mennonite ancestry. The available Y chromosome data from those Mennonite samples is available here. I have been looking forward to seeing the release of the SMGF's autosomal and X chromosome databases for many years. Now it would appear that this data won't be released anytime in the near future. 

Due to concerns I had about the future of the SMGF, I e-mailed my contact Ali Nelson there on May 31. I received her response on June 5 as below:

"Due to Ancestry.com's acquisition of GeneTree and SMGF, we are no longer updating the database. That will be up to Ancestry to decide how and when to do updates and I don't have any more information than that. SMGF is also not running any more samples - everything now belongs to Ancestry so it is up to them to run the remaining samples. We are winding things down here in the office and actually will not exist for much longer so I don't see any reason for you to send us an updated copy of the Grandma database. All of our assets now belong to Ancestry.com and they are making all of the decisions from here on out. You can contact Ancestry.com directly and hopefully they can give you more details. They have an email address devoted to questions regarding SMGF so you can try contacting them there first - it is smgfsupport@ancestry.com."

On June 21 Ali Nelson sent additional information as follows: 

"In regard to the SMGF website, it is still going to remain functional.  In fact, we will be doing an update in the next week or two so a lot of new samples will go online."  

Ali also said that the SMGF will be officially closing its doors on or about July 13.

The information provided by Ali Nelson in her responses are disappointing in a number of ways as follows:
1. She confirmed that SMGF is closing its doors and will not be operating much longer.
2. The SMGF's Y chromosome and mitochondrial DNA databases will be updated within the next several weeks for the very last time.  When and if Ancestry.com decides to do additional updates to the SMGF databases remains to be seen.
3. It is unclear when, if ever, additional testing of samples currently at the SMGF will be carried out.  There are thousands of samples at the SMGF for whom no Y chromosome or mitochondrial DNA results have been released.

On the positive side, the SMGF will soon be releasing new data for many people whose data hasn't previously been released. It is also good that the SMGF DNA samples are still safe and sound and are under the watchful care of a large relatively stable company. Hopefully Ancestry.com will provide funds for testing the samples for which no Y chromosome or mitochondrial DNA results are available and will also update the SMGF Y chromosome and mitochondrial DNA databases from time to time. I would also be grateful if Ancestry.com eventually releases the SMGF's autosomal and X chromosome databases. While it is true that the use of large autosomal SNP arrays has largely supplanted the testing of autosomal STRs, the release of the SMGF's autosomal STR database would be of significant interest and benefit to the genetic genealogy community. In my opinion the demise of the SMGF is a significant loss to the entire genetic genealogy community. The SMGF had an altruistic vision to create an integrated genealogy database with correlated genetic data that would be accessible to the general public. I remain hopeful that something good will come from this and that Ancestry.com will pick up the mission where the SMGF left off.

"Finding Your Roots with Henry Louis Gates, Jr." - DNA in the Eighth Episode


Featured last night on the eighth episode of Finding Your Roots with Henry Louis Gates, Jr. were Martha Stewart, Margaret Cho and Dr. Sanjay Gupta. Each of these "trailblazers in media" descend from very recent immigrants to the United States and thus face special challenges in tracing their family trees. The good news is that I was granted the reprieve I hoped for last week and the bad news is that I was granted the reprieve I hoped for last week. There was very little genetic genealogy in this episode, so this will be a relatively quick and easy review.

Early on in Martha's segment after discovering that many of the branches of her family tree were populated with ancestors who were "highly skilled in the domestic arts" - like basketweavers, iron workers and seamstresses, Dr. Gates commented that this "makes you believe in genetic predeterminism," the idea that an individual's development is preordained by heredity. Later, Martha mentioned her dog is named "Genghis Khan" after Dr. Gates' team linked her ancestry to the Tatar people who were subjugated under Khan and they laughingly discussed the possibility that she could be descended from him. (Filed under "things that make you go 'Hmmm'".)

The DNA portion of the show wasn't until the very end of the episode (starting at 46:40) and started with an analysis by  23andMe of Martha's direct maternal line DNA (mtDNA). (Mitochondrial Haplogroups are included with 23andMe's genome scan.) This ancestral line had been traced back by Dr. Gates' genealogy team to a maternal ancestor with the last name Albiniak who lived in the town of Janów, which apparently implies that the family was Muslim. Martha's mtDNA haplogroup was determined to be W6, a Central Asian haplogroup which, according to Dr. Gates, spread to the area around modern day Pakistan, thus supporting his genealogists' conclusion regarding a potential Islamic heritage for Martha's direct maternal line (mother's mother's mother's mother, etc...). Although this is quite an unexpected and intriguing result for someone who seems as WASPy American as apple pie, Martha didn't appear to be particularly surprised by the news.

A website dedicated to the W haplogroup states that W6 reaches its highest concentrations in Georgia at 5.2% compared to 2.7% in Pakistan. Wikipedia states that Haplogroup W, not the subclade of W6, is found in Pakistan. I was unable to find enough information on this relatively rare haplogroup to reach any definite conclusions on its relevancy to Martha's family history.

Next, Dr. Gates revealed Sanjay's mitochondrial DNA haplogroup to be U2c, which he said occurs most often in the Sindhi people of Southern Pakistan. This result is not surprising for a person of Indian heritage like Sanjay and coincided with his mother's family's recent migration (1947) from the area that is now Pakistan. This results suggested that Sanjay's direct maternal ancestral line was likely in this region for thousands of years. It would have been neat to find out what his paternal line haplogroup (Y-DNA Haplogroup) was too, but Dr. Gates chose to focus on mtDNA exclusively in this episode.

Margaret was in for a bit of surprise when her mtDNA haplogroup was reported as D5a2a1b. This haplogroup reaches highest concentrations in China, not Margaret's family's homeland of Korea. In fact, D5a is extremely rare in Korea, appearing in less than one percent of the population. Further, Dr. Gates mentioned that "another test" showed that her paternal haplogroup also traces back to China instead of Korea. This other test would have been a Y-DNA Haplogroup test. Since Margaret herself does not have a Y-Chromosome, to reach this conclusion, Dr. Gates team must have tested her father or a brother. Unfortunately, the specific Y-DNA Haplogroup was not revealed. I would have liked to see it if only for the reason that, in my experience, Asian DNA is not widely discussed and would benefit from more visibility in our research and discussions.

Even though mtDNA haplogroups were the only DNA results explored in this episode, it was very interesting because each of the guests had a relatively rare haplogroup, at least as far as what exists in our databases at this point in time. (I searched the list of about 1000 people that I am sharing with at 23andMe and none of them had any of the three mtDNA haplogroups mentioned here. I also searched the corresponding haplogroup projects at FTDNA and found very few from these subclades.) Haplogroups are indicative of deep ancestry and are not generally genealogically relevant, however when the documentary paper trail runs out, they can provide clues as to the family's historic location and migratory pattern.  Dr. Gates explains it like this, "Certain DNA tests act like a genetic global positioning system, providing valuable clues about where our ancestors once lived." Apparently Dr. Gates' team used 23andMe's genetic scans to determine these haplogroups. This is a little surprising since haplogroup research is actually more of a specialty of Family Tree DNA, the other DNA testing company often featured in the series. 23andMe includes this information with their autosomal DNA test, but offers no further information beyond the haplogroup designation. At Family Tree DNA, a person can get the detailed list of the exact STRs (Y-DNA) and mutations (mtDNA), thus enabling matching with other individuals in the large FTDNA databases containing these specific results. (I suggest the Y-DNA 37 marker test and the mtDNA Plus.)

On my wish list would have been admixture breakdowns for the guests (remember those pie charts in earlier episodes?), especially Martha and Sanjay. Martha's results could have been very interesting if any of her Tatar autosomal DNA remains. I'm not sure what her breakdown would look like, but I imagine there would be an Asian component. Since her maternal grandmother was an Albiniak and her genetic contribution makes up about 25% of Martha's genome, it is certainly plausible that some autosomal genetic traces of her Tatar ancestors remain. (I note from an entry in Wikipedia that the Muslim Tatars were allowed the unusual right of intermarriage with Catholics in Poland, which may help to account for that portion of Martha's family tree.)

I would have also liked to see a breakdown of Sanjay's genetic ancestral origins to discover if, as a South Asian Indian, he has substantial admixture (of both Asian and European autosomal DNA) or primarily just Asian DNA. It is always interesting to me to see how these two components balance out. Margaret's autosomal DNA likely wouldn't have revealed anything significant since the major testing companies have yet to commercially analyze East Asian DNA in any great detail, but you never know.

I also really missed seeing the familiar faces from the two DNA testing companies regularly featured in this series.

Next week we will delve back into the genetics of African Americans with guests John Legend and Wanda Sykes who are both descended from free people of color.  Now that I've had my break, I hope this episode is packed full of DNA research on these intriguing family trees, especially since there are only two episodes left!

"Finding Your Roots with Henry Louis Gates, Jr." - DNA in the Sixth Episode


The sixth episode of Finding Your Roots with Henry Louis Gates, Jr., investigating the mixed ancestry of Robert Downey, Jr and Maggie Gyllenhaal, aired last night on PBS. These two actors not only share a profession, but both have a mixture of Eastern European Jewish ancestry as well as deep American roots. Because of these commonalities, I thought their stories worked well together. I was thoroughly entertained by this episode for three reasons: 1) I love Robert Downey, Jr., 2) I discovered that Maggie is my cousin through our shared 10th great grandfather John Lothrop and 3) The appearance of Bennett Greenspan, President of Family Tree DNA (or as Professor Gates called it "FamilyTreeDNA.com", which I am sure was much appreciated).

Unfortunately, the DNA portion was only about five minutes long again this week (starting at 43:00), nevertheless it felt a bit more substantial to me than last time. I was happy to see a number of different tests featured: 23andMe's Ancestry Painting, FTDNA's Population Finder and mtDNA testing.

First, Robert Downey, Jr., whose father is half Jewish, was presented with a pie chart of his 23andMe Ancestry Painting, which breaks down a person's genetic ancestral origins into three populations - European, African and Asian. (Note - this test is included in the 23andMe Personal Genome Service and is presented as a chromosomal chart rather than a pie chart.) Dr. Gates has repeatedly stated that this test reveals "European, African and Native American ancestry", however I would like to remind readers that the Asian reference population simply "stands in" for Native American. 23andMe does this because the genetic makeup of the two populations is similar, but in reality, this is not quite the same as having reference samples from Native Americans and the two should not be confused with each other. For instance, a person could show Asian DNA on their Ancestry Painting, but one should not automatically reach the conclusion that they have Native American ancestry because they may have ancestors from China, Japan, Korea or any other Asian region. If a person has reason to suspect that they possess Native American ancestors and knows of no other Asian ancestry, then this test is useful for that purpose. As is typical of most people, RDJ guessed that his ancestry would be more "interesting" than it was and seemed disappointed to learn that his Ancestry Painting showed 100% European ancestry. As I have mentioned before, this actually encompasses all of Europe, including the Middle East, so it doesn't rule out RDJ's Jewish ancestry.

Next, Dr. Gates presented RDJ with what I believe was his Population Finder admixture from Family Tree DNA. (The PopFinder feature is included with the Family Finder test.) This test broke down RDJ's ancestral origins a bit more, showing that he is about 20% Middle Eastern or Jewish.  That is slightly under what we might expect for him with a father who is half Jewish, so his one Jewish grandparent may have had a little something else mixed into his/her ancestral origins.

Later, Dr. Gates sat down with Maggie to discuss her mitochondrial DNA (mtDNA) which is inherited straight down the maternal line, passed from mother to child as far back "as you can think" enthused Maggie. Maggie's mom is Jewish, so it makes perfect sense that Maggie's mtDNA has been inherited from one of four Jewish founding mothers who lived thousands of years ago. The voiceover explained, "Scientists have concluded that 40% of Eastern European Jews share genetic lineages that reach back two to three thousand years, remarkably to just four women...today about 3.5 million people descend from one of these four maternal ancestors." Haplogroup Project Administrator and genetic genealogist Vince Tilroe points out that Bennett Greenspan and Professor Gates were most likely speaking of mtDNA Haplogroups K1a1b1a, K1a9, K2a2a, and N1b when referring to the four Jewish maternal lines. These were identified in the 2006 study by Dr. Behar, "The Matrilineal Ancestry of Ashkenazi Jewry: Portrait of a Recent Founder Event".

Bennett further explained that Maggie had "hit the jackpot" with about 150 exact matches in Family Tree DNA's mtDNA database.  Bill Hurst, Administrator of the mtDNA Haplogroup K Project and a leading expert on this haplogroup, theorizes that if Maggie's mtDNA haplogroup is K, she likely falls into the K1a1b1a ancestral haplotype. Hurst explains that this haplotype's match numbers most closely coincide with the those that Bennett cited from FTDNA's database. At the HRV1/HRV2 level of testing, this haplotype now has 194 exact matches, but at the time of filming may have had closer to 150. He also notes that K2a2a is now called K2a2a1 due to recent discoveries. (There are three levels of mtDNA testing offered by Family Tree DNA which you can read about here.)

At the end of the episode Professor Gates discussed the question of "nature versus nurture" with his guests and asked Robert Downey, Jr. if he feels that his ancestral past has shaped who he is today. In answer, RDJ insightfully commented, "I don't see how it couldn't...it's informing you whether you're conscious of it or not, so probably better to be conscious." The episode ended with RDJ expressing his feelings about this experience with Dr. Gates, explaining that he felt that this was just the beginning of his journey of self-discovery.

I hope this show has also inspired all of you to begin (or continue) the journey into your ancestral roots. The television listing says that the show will be using DNA to "determine which parts of Africa their ancestors came from." That sounds promising!
